Cross Compatibility, Seed Germinability, and Embryo Rescue following 2x * 4x Crosses in Hibiscus syriacus




Abstract
To obtain basic data for breeding triploid cultivars in Hibiscus syriacus L., crossability, germinability of hybrid seeds, embryo rescue, and relationship between seed weight and LEA (late embryogenessis abundant) protein were examined after crosses between two tetraploid cultivars and 48 diploid cultivars in Hibiscus syriacus L.. The crossability of 2x ¡¿ 4x crosses ranged from 0 to 100%, regardless male parents. Hybrid seeds of each combination showed continuous distribution frequency in the weights of normal, shrunken, and aborted seeds. Normal seeds with weighing more than 18 §· germinated above 90%, but germinability of the shrunken seeds decreased remarkably. The shrunken seeds contained immature or abnormal embryos in various sizes in which LEA proteins were accumulated at lower level. Relatively healthy embryos, more than 15 §· in their fresh weight, excised from the seeds of 40-45 days after pollination germinated above 90% under non-aseptic condition of peat-perlite medium when treated with 200 ppm NAA for two hours. More than 90% of smaller embryos were able to rescued by aseptic culture on MS basal medium supplemented with 0.01 §·/L of GA©ý and NAA.
